ABSTRACT

@#To study the protective effects of Roudoukou-8 San extract on hypoxia/reoxygenation injury of cardiac myocytes and its relationship with tyrosine protein kinase 2(JAK2)/signal transducer and activator 3(STAT3)signaling pathway, hypoxia/reoxygenation injury model of H9c2 cells was built by sodium dithionite(Na2S2O4). The vitality of the cells was tested by CCK-8; the contents of lactate dehydrogenase(LDH), creatine phosphate kinase(CK)and aspartate aminotransferase(AST)in cell culture medium were tested by fully automatic biochemical analyzer; the contents of catalase(CAT), superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)in cells were tested by kits; cell apoptosis degree was tested by hoechst staining. And the protein expressions of JAK2, p-JAK2, STAT3, p-STAT3, Bcl-2 and Bax in myocardial cells of each group were tested by Western blot. Hypoxia for 1 hour and reoxygenation for 3 hours of 2 mmol/L Na2S2O4 caused damage of about 50% H9c2 cells. Compared with the model group, the extracts of Roudoukou-8 San with different concentrations could increase the viability of cells. Besides, contents of CK, LDH and AST in cell culture medium were decreased significantly, while contents of CAT, SOD and GSH-Px were increased significantly. At the same time, the expression of p-JAK2, p-STAT3 and Bcl-2 were significantly increased and that of Bax was significantly decreased. The effects of Roudoukou-8 San extract could bereduced by AG490 blocker. Therefore, Roudoukou-8 San extract possesses protective effects on Na2S2O4 induced-hypoxia/reoxygenation injury of cardiac myocytes through JAK2/STAT3 signaling pathway.

ABSTRACT

Objective To evaluate the nutrional status and malnutrition risks of hospitalized children with acute lymphoblastic leukemia (ALL),provide nutrition support for subsequent treatment,lower nutritional risks and improve therapeutic effects;to complete the statistical analysis of height and weight across age through data normalization.Methods A total of 592 children diagnosed ALL from August 2014 to September 2016 at Beijing Children's Hospital were enrolled,and the directly measured height and weight which reflected the nutritional status of children were collected and normalized.Nutritional risk screening was completed by STAMP screening tool and the nutritional status was evaluated by three Z scores HAZ<-2,WAZ<-2 and WHZ<-2.The effects of nutrition intervention were investigated by weight,hemoglobin and albumin changes between before and after L-asp treatment.Results The Z score test showed that there was no significant difference between the nutritional status of pretreatment ALL patients (0.34% growth retardation,2.36% lower body weight,0.17% emaciation) and normal healthy children (3% malnutrition);after comparing the STAMP nutritional risk assessment and t test results for children with ALL,it was found that the proportion of children with high malnutrition risk (score≥4) was 9.71%,which had a strong correlation with body mass index;the children with body mass index less than 0.80 (50% children having malnutrition) or greater than 1.30 (20% children having malnutrition) had a higher risk of malnutrition,and the body mass indexof the high-risk group (0.852 kg/m2) were significantly lower than that of the low risk group (1.051 kg/m2,score ≤ 3);there were significant differences in height and weight between children with STAMP ≤ 3 and STAMP ≥ 4 (all P <0.05) Nutritional interventions made no significant weight change during treatment;For children with STAMP ≤3 and STAMP ≥4 in L-asp treatment,the hemoglobin value changed from (87.46± 19.27) g/L to (95.12±13.51) g/L and (101.55±21.97) g/L to (95.05±11.22) g/L respectively (all P=0.001);The albumin of children with STAMP≤3 changed from (40.63±4.149) g/L to (41.20±5.266) g/L in treatment and that of children with STAMP ≥4 changed from (40.96±8.429) g/L to (42.17±3.574) g/L in treatment (P=0.20,0.05).Conclusion There are no obvious indications of malnutrition in children with ALL,but nearly 10 % of them have malnutrition risks.Special dietary guidance is needed during the treatment.Nutrition intervention is effective in the treatment of L-asp.Normalized data is more convenient to use in statistical analysis with more accurate results,which can be used as a supplementary method for cross-age group statistical analysis of children's height,weight and other parameters.

ABSTRACT

Objective To evaluate the value of color flow angio(CFA)in judging the status of thyroid function by analyzing the ultrasonographic features and quantitatively measuring the blood flow signals in patients with thyroid diffuse lesions.Methods Totally 100 patients with thyroid diffuse lesions diagnosed by ultrasonography between Oct.2016 and Jun.2017 were divided into three groups:hyperthyroidism group(n=30),hypothyroidism group(n=34)and euthyroid group(n=36).Moreover,30 cases of healthy volunteers were chosen as control group.The blood flow index including vascularity index(VI)value and Vascularity value of region of interest from all cases was measured through CFA imaging technique.The differences of VI and Vascularity from above 4 groups were compared using one-way analysis of variance.Comparison among groups was carried out by SNK-q test.The subjects' work curves(ROC curves)were plotted and analyzed using clinical diagnosis as gold standard.At the same time,the correlation between VI and thyroid function in hyperthyroid group and hypothyroidism group was analyzed through Pearson correlation coefficient.Results The levels of VI and Vascularity in hyperthyroid group,hypothyroid group,euthyroid group and control group were decreased gradually with significant differences between every two groups.VI : control vs hyperthyroid,hypothyroid and euthyroid group: q=13.67,7.00 and 3.93,all P<0.01.Euthyroid vs hyperthyroidism and hypothyroid group: q=10.35,P<0.01 and q=3.27,P<0.05.Hyperthyroidism vs hypothyroid group: q=7.09,P<0.01.Vascularity: control vs hyperthyroid,hypothyroid and euthyroid group: q=15.23,10.16 and 6.58,all P<0.01.Euthyroid vs hyperthyroidism and hypothyroid group: q=9.33 and 3.83,both P<0.01.Hyperthyroidism vs hypothyroid group: q=5.55,P<0.01.The area under the curve of hyperthyroidism group and hypothyroidism group was 0.733,with 9.526%as the diagnostic cutoff point.The sensitivity and specificity were 70.0%and 76.5%respectively.The area under the curve of the abnormal group and normal group was 0.768,with 7.404%as the diagnostic cutoff point.The sensitivity and specificity were 62.5%and 88.9%,respectively.The VI value of hyperthyroidism group was positively correlated with FT3 and FT4(r=0.584,0.499,both P<0.05),and negatively correlated with TSH(r=-0.447,P<0.05).The VI value of hypothyroidism group was negatively correlated with FT4(r=-0.342,P<0.05),and had no significant correlation with FT3 and TSH(r=0.121,-0.007,P>0.05).Conclusion CFA imaging technology can quantitatively measure the blood flow signals of thyroid parenchyma and evaluate the thyroid function in patients with thyroid diffuse lesions.

ABSTRACT

Objective To explore the value of contrast-enhanced ultrasound (CEUS) scores, ultrasonic elastography (UE) and their combined scores in diagnosing breast lesions of breast imaging reporting and data system (BI-RADS) 3-4. Methods From October 2015 to March 2016, 60 BI-RADS 3-4 solid breast masses diagnosed by conventional ultrasound underwent UE and CEUS. According to the relevant indicators, all cases were scored and proved by surgical pathology. The diagnostic sensitivity, specificity and accuracy of CEUS scores, UE and their combined scores were evaluated, and the receiver operating characteristic (ROC) curve was drawn. Results Twenty-nine benign and 31 malignant lesions were confirmed by pathology. The area under the ROC curve of CEUS combined with UE scores was 0.971 which was higher than that of CEUS Scores and UE alone (0.916 and 0.908, respectively). Moreover, the sensitivity, specificity and accuracy were 87.1%, 96.6% and 92.0% for CEUS combined with UE Scores, 77.4%, 93.1% and 85.0% for CEUS Scores, 74.2%, 93.1% and 83.3% for UE, respectively. Conclusion CEUS combined with UE scores can improve the differetial diagnostic accuracy for BI-RADS 3-4 breast lesions.

ABSTRACT

ObjectiveTo study the feasibility and stability of hepatocellular carcinoma model established in mouse with the guidance of ultrasound, and to explore the value of ultrasonography in hepatocellular carcinoma model.MethodsThe type of H22 cells, whose concentration was 2×107/ml, were injected into the left liver of 30 mice with the guidance of ultrasound. After injection, the mice were examined by ultrasound on the tenth and twentieth day, and implemented laparotomy on the twentieth day. The tumors were examined pathologically. The formation rate and growth of tumors were evaluated, and the ultrasonic features of tumors were observed.ResultsThe formation rate of implanted tumors was 96.7% (29/30), and the peritumoral blood flow could be detected. The diameters measured by ultrasound and explorative laparotomy were (8.4±0.3) mm and (8.6±0.2) mm, respectively (P>0.05). The pathological examination showed that the tumors were hepatocellular carcinoma.ConclusionEstablishing mouse model of hepatocellular carcinoma by ultrasound-guided is simple and minimally invasive, and it provides an ideal model of high formation rate for the study of hepatocellular carcinoma.

ABSTRACT

Objective To evaluate the prognostic factors of hepatocellular carcinoma (HCC) patients with treatment of ultrasound-guided pereutaneous radiofrequency ablation (RFA).Methods A total of 104 patients with 147 HCC tumors who had 36-month regular follow-up after percuatenous RFA therapy in the First Affi liated Hospital of Harbin Medical University between May 2008 tand June 2012 were included in this study. Contrast-enhanced ultrasound (CEUS) was the main method in evaluating the therapeutic effect of RFA. Kaplan-Meier model and Log-rank test were used in univariate analysis, and Cox regression model was used in multivariate analysis to identify the independent factors on therapeutic effect of RFA.Results The 0.5-, 1-, 2-, 3-year overall survival rates after RFA were 96.1%, 92.0%, 80.0%, 53.3% respectively, and the mediansurvival time was 19 months. 1-year local recurrence rate was 13.6%. The univariate analysis showed that Child-Pugh classifi cation, original lesion condition, intrahepatic recurrence, ablative area, and RFA priority were related to survival rate (χ2=7.119,P=0.008;χ2=20.485,P=0.000;χ2=14.160,P=0.000;χ2=11.825, P=0.001;χ2=7.979,P=0.005). The Child-Pugh classifi cation, original lesion condition, intrahepatic recurrence were identified as independent prognostic factors of survival rate (P=0.001;P=0.005;P=0.003). The univariate analysis showed that virus marker, original lesion condition, hepatic background, tumor diameter, tumor boundary, relation with adjacent organ and vessels, ablative area, RFA priority weresignifi cantly related to local recurrence rate (χ2=7.234,P=0.007;χ2=9.083,P=0.003;χ2=7.791,P=0.005;χ2=13.042,P=0.000;χ2=20.657,P=0.000;χ2=58.615,P=0.000;χ2=6.681,P=0.010). The ablative area and tumor boundary were identified as independent prognostic factors of local recurrence rate (P=0.017;P=0.002).Conclusions Child-Pugh classifi cation, original lesion condition, intrahepatic recurrence were identifi ed as independent prognostic factors of survival. And ablative area and tumor boundary were identified as independent prognostic factors of local rcurrence rate. In order to improve patient`s survival and reduce local recurrence in high-risk cases, it is importantant to establish appropriate follow-up plan and multidisciplinary cooperation framework within a standard treatment procedure.

ABSTRACT

ObjectiveTo compare the clinical value of contrast-enhanced ultrasonography (CEUS) and conventional ultrasonography in diagnosing polypoid lesion of gallbladder (PLG).MethodsThe postoperative pathology and clinical manifestation of 49 PLG patients were retrospectively analyzed in the First Afifliated Hospital of Harbin Medical University. Of them, 7 cases were maligant lesions, 4 cases were adenomas, 24 cases were polyps of gallbladder and 14 cases were adenomyomatosis of gallbladder. With postoperative pathology as golden standard, the sensitivity, speciifcity and PLG speciifcity were calculatedfor conventional ultrasonography, CEUS and conventional ultrasonography plus CEUS.ResultsThe sensitivity, specificity and PLG specificity were 33.3%, 74.3% and 68.3% for conventional ultrasonography, and 66.7%, 85.7% and 82.9% for CEUS respectively. The conventional ultrasonography plus CEUS were 83.3%, 88.6% and 87.8%.ConclusionCEUS could signiifcantly improve the diagnostic accuracy.

ABSTRACT

Objective To assess the value of imaging examination technologies such as 2-D ultrasound(2-DUS),helical computed tomography (CT),contrast-enhanced ultrasonography (CEUS) and contrast-enhanced CT (CECT) in the positioning and qualitative diagnosis of pancreatic diseases.Methods 2-DUS,helical CT findings were comparatively reviewed to analyze in 44 cases(45 lesions) with pancreatic space-occupying lesions verified by operation and pathology,and investigated the enhanced mode and the perfusion features of CEUS and CECT in different phases.Results In 45 lesions,there were 29 malignant lesions and 16 benign lesions.CEUS and CECT had greater diagnostic value compared to 2-DUS and helical CT.The sensitivity,specificity and accuracy of 2-DUS for the malignant lesions were 72.41 %,62.50% and 66.67 %,while the helical CT were 68.97%,75.00 % and 68.89 %.The sensitivity,specificity and accuracy of CEUS for the malignant lesions were 93.10%,87.50% and 91.11%,while the CECT were 86.21%,93.75% and 86.67%.There was no statistical difference between CEUS and CECT in detecting pancreatic space-occupying lesions.Conclusions CEUS and CECT can provide more accurate diagnosis on for pancreatic space-occupying lesions than 2-DUS and helical CT.Following CECT,CEUS is a new technology for diagnosis on pancreatic space-occupying lesions.

ABSTRACT

Objective To provide the evidence for the clinical prevention and early treatment for the complication by testing microcirculation of the middle fingertip in renal failure patients with hemodialysis.Methods High-frequency ultrasounds combined with E-flow technique were applied to detect the Doppler velocitg index of middle fingertip's nail bed arteriole and ventral arteriole in 35 renal failure patients with hemodialysis.Peak systolic velocity(PSV),end-diastolic velocity(EDV)and vascular resistance index(RI)were separately measured,and comparison were made with healthy adults and renal failure patients without hemodialysis(sex and age matched).Results Compared with normal control,PSV and EDV of middle fingertip's nail bed arteriole and ventral arteriole were degraded statistically(P<0.05),while RI was increased statistically(P<0.05)in renal failure patients with and without hemodialysis.Compared with renal failure patients without hemodialysis,PSV and EDV were degraded(P<0.05)and RI was increased(P<0.05)in renal failure patients with hemodialysis.Conclusions High-frequency ultrasound combined with E-flow imaging technique can evaluate the microvascular peripheral resistance in renal failure patients with hemodialysis.

ABSTRACT

Objective To investigate the value and the enhanced mode of time-intensity curve(TIC)combined with gamma curve of contrast-enhanced ultrasound in diagnosis of hepatocellular carcinoma.Methods The ultrasonic contrast images of 34 patients with hepatocellular carcinoma by the gamma curve were obtained by the time before,increased branch slope parameter, curve sharpness, gamma modulus, peak time,peak intensity, the average pre-peak intensity, increased branch slope and pre-peak area.Results Compared with the peripheral hepatic parenchyma, the curve of the tumor tissue ascended as well as declined more rapidly, peaked earlier with a more intensive peak value.There were significant differences between the two groups in all the data mentioned-above (P<0.05 ).Conclusions TIC and gamma fit quantitative analysis can reflect the different perfusion characteristics between hepatocellular carcinoma and normal hepatic parenchyma objectively and accurately,so as to provide the quantitative criterion and basis for the diagnosis and hemodynamic study of the hepatocellular carcinoma.

ABSTRACT

objective To assess relationship between microvascular changes of hemodynamics of the fingertip and evolution of hypertension in patients with essential hypertension(EH)by enhanced power Doppler imaging(E-Flow).Methods The right ring fingertip'S blood flow peak systolic velocity(PSV),end-diastolic velocity(EDV),resistance index(RI)of finger ventral arteriole and nail bed arteriole in thirty-nine patients with EH were separately observed and were made contrast with 35 age-matched healthy subjects by E-Flow imaging.Results Compared with traditional colour Doppler flow imaging and power Doppler imaging,E-Flow imaging was clearer to demonstrate the distribution and movement of the fingertip'S microvascular.It was shown that the PSV and EDV in the ring finger ventral arteriole and nail bed arteriole of EH were decreased(P<0.05),but the RI increased(P<0.05),along with the evolution of EH.Conclusions Fingertip'S microvascular changes of hemodynamics could be a criterion to detect obstruction alternations of essential hypertension.E-Flow imaging iS superior to traditional color Doppler flow imaging and power Doppler imaging in differentiating microvascular and could provide a new method in assessing microvacular changes of patients with EH for clinic.
